Berättelsen om två rörmokare från Brooklyn - bröderna tillika bästa vännerna Mario och Luigi. Mario är den modige med lets-a-go-inställning och Luigi är den ständigt ängslige som föredrar att inte gå nånstans. Filmen börjar med att bröderna gör upp räkningen med sin krisande rörmokarfirma och tar oss med på ett virvlande äventyr genom Svampriket.
